Stabilization of protein-latex complexes at high ionic strength

چکیده

When studying the colloidal stability of an immunolatex formed by F(ab')2 protein fragments and a polystyrenepolychloromethylstyrene copolymer, striking results were obtained. After the expected particle aggregation induced by an increase in the electrolyte concentration (KBr), subsequent restabilization occurred when this electrolyte concentration was increased even more. This non-DLVO behaviour was studied under different experimental conditions (ionic strength, pH and surface coverage) and with a different solid support (polystyrene) and covering protein (bovine serum albumin) in order to generalize to other systems. An explanation based on ion hydration layer, similar to that proposed for hydrophilic surfaces, is assumed.

Colloidal stability of IgG- and IgY-coated latex microspheres.

The stabilization of antibody-latex complexes at high salt concentration is an event that cannot be explained by the widespread DLVO theory. Adsorption of antibodies on polystyrene latex usually leads to a loss in colloidal stability. However, after the expected particle aggregation induced by an increase in ionic strength, an 'anomalous' restabilization occurs when the electrolyte concentratio...

Controlled Release of Proteins Bound to Spherical Polyelectrolyte Brushes

We discuss the interaction of proteins dissolved in aqueous solution with spherical polyelectrolyte brushes (SPB). The SPB consist of a solid core particle of colloidal dimensions (ca. 100 nm in diameter) onto which long polyelectrolyte chains have been grafted. Immersed in aqueous solution of proteins these SPB will take up high amounts of protein if the ionic strength is low. At high ionic st...

Differences between chemisorbed and physisorbed biomolecules on particle deposition to hydrophobic surfaces.

This study examines differences between chemisorbed and physisorbed biomolecules on bacterial adhesion to both hydrophobic and hydrophilic surfaces that are biologically nonspecific. Bacteria-sized latex microspheres were used as a simplified model in order to study these factors that affect microbial adhesion. Two biomolecules (protein A, poly-D-lysine) were covalently bound to microspheres in...

On the restabilization of protein-covered latex colloids at high ionic strengths.

Recent experiments on restabilization of protein-covered latex colloids at high ionic strengths reported by Lopez-Leon et al.(1) revealed strong specific anion effects. The same authors also emphasized that a recent polarization model, which involves both hydration and double layer forces, can account only for some of their experimental results but are in disagreement with other experimental re...

Structure and Transport of Latex Microgels in Aqueous Suspension

Cationic latex microgel particles were studied in dilute aqueous suspensions of varying ionic strengths. Static light scattering indicates a Gaussian distribution of polymer density over the length scales probed. The measured radius of gyration is up to 50% larger than that obtained from TEM measurements of dried precursor particles. This radius of gyration is observed to be weak function of io...
